锦鸡儿属植物的遗传多样性及其种间关系

Genetic Diversity and Relationships among Species of Caragana Fabr.

摘要: 利用RAPD技术,对甘肃省自然分布的11种锦鸡儿属(CaraganaFabr.)植物的遗传多样性及其种间关系进行了研究。从34个10碱基随机引物中筛选出能产生稳定多态性标记的引物11个,共扩增出128个位点,主要集中在200~2000bp之间,其中104个表现出多态性,多态性比率高达81.25%;根据POPGENE32软件计算的Shannon多样性指数与Nei指数也较高,分别为0.3889和0.2517。说明锦鸡儿属植物具有丰富的遗传多样性,对环境变化的适应能力较强。应用UPGMA法进行聚类分析,结果表明,RAPD分析所反映的种间遗传关系与传统的形态学分类结果基本一致,但也有个别种的从属地位稍有变化。这说明还需要将分子标记的研究结果与形态学和其他方法结合起来进一步进行研究。

Abstract: RAPD markers were used to assess the genetic diversity of Caragana Fabr. and to estimate genetic relationships among the eleven species growing in Gansu province. The 11 primers were screened from 34 10-bp arbitrary primers, and a total of 128 loci were amplified; the length of fragments was between 200 bp and 2 000 bp, among which, 104(81.25) were polymorphic. The Shannon's information index and Nei s gene diversity were 0.3889 and 0.2517, respectively. The results showed that the Caragana Fabr. had a high level genetic diversity, and its adaptability to environmental change was strong. A dendrogram based on genetic distances was constructed by UPGMA method of POPGENE version 1.32. The conclusion was almost consistent with the results of traditional morphologic taxa, except hypotaxis status of very few species was slightly changed. It indicated that it is necessary for further studying their relative relations to combine molecular markers with morphologic and other methods.
