Effects of Forchlorfenuron on Callus Induction and Proliferation of Reaumuria soongorica and Its Formula Optimization by Response Surface Method

Abstract  One-year old stem of Reaumuria soongorica was used as the explant to study callus induction and proliferation of desert plant Reaumuria soongorica by different CPPU, 6-BA and NAA concentrations. Results indicate that callus is well induced on 1/2 MS agar medium supplemented with 0.1 mg·L-1 CPPU and 1 mg·L-1 NAA. According to the principles of Response Surface Method, 2 factors and 5 levels are adopted in central composite design experiment. The experiment shows that optimal condition for callus proliferation is 1/2 MS agar medium with 0.1195 mg·L-1 CPPU and 1.1 mg·L-1 NAA. Under this condition, the callus proliferation rate is up to 4.7414 and it is 99.15% of the predictive value. The optimal period of the secondary culture of the calluses is 30 days. The research suggests that callus proliferation medium can be optimized by the Response Surface Method, and the proliferation rate can be increased, so the new method is effective and economical.
