荒漠植物红砂愈伤组织诱导与增殖研究

Induction and Proliferation of Callus in Desert Plant Reaumuria soongorica

Shapotou Desert Research & Experimental Station, Cold and Arid Regions Environmental and Engineering Research Institute, Chinese Academy of Sciences, Lanzhou 730000, China Abstract: Leaf and young stem were used as the explant to study callus induction and proliferation in desert plant Reaumuria soongorica. The results indicated that callus was well induced from tender shoot tips on MS agar medium supplemented with 2\^0 mg\5L-1 2,4-D and 0\^2 mg\5L-1 6-BA, pH at 5\^8, and the induction rate reached as high as 100%. The induction rate was lower in leaf than that in stem. The optimal conditions for callus proliferation were on the MS agar medium with 1\^0 mg\5L-1 2,4-D and 0\^1 mg\5L-1 6-BA, and the callus kept their incompact structure on such medium. Callus on media with different NaCl levels was found that it grew quickly in 50mM NaCl, but it was died in higher than 300 mM NaCl.

摘要: 以红砂的叶和嫩茎为外植体,进行愈伤组织诱导与增殖研究。结果表明: 红砂的嫩茎是诱导愈伤组织最快的外植体; 其愈伤组织诱导的最适培养基为: MS+6-BA(0.2 mg·L-1+2,4-D(2.0 mg·L-1),pH值5.8,诱导率可达100%; 而叶片的诱导相对要慢。愈伤组织的增殖在MS+6-BA(0.1 mg·L-1)+2,4-D(1.0 mg·L-1) 的培养基中最为理想,并维持其良好的组织结构, 其最适继代周期为30~40 d。愈伤组织在含50 mM NaCl的培养基
上生长速度最快,但NaCl含量大于300 mM的培养基上致死,不再生长。
