Abstract We introduced a better method which do best for the purity and concentration of total DNA of seven Calligonum species: Calligonum mongolicum Turcz. , C. junceum (Fisch. et Mey.) Endl., C. aphyllum (Pall.) G rke,C. rubicundum Bge., C. cordatum E. Kor. ex N. Pavl., C. densum Borszcz., C. arborescens Litv.. This method is an improved one upon material and method based on the traditional CTAB method. The result showed that it could effectively eliminate the affection of secondary materials to extracted total DNA from Calligonum, and is suitable for PCR of ISSR and other genetic research. Also, it provides a reliable experimental method for relevant researches on Calligonum.
