一种生物土壤结皮中蓝细菌(Microcoleus spp.)的分子鉴定

Isolation and Identification of Synechococcus from Biological Soil Crust

摘要: 在从北京怀柔风沙防治技术示范区采集的沙样中,采用96孔板有限稀释法分离出的微生物,形态学初步鉴定分别为蓝细菌。进行PCR和测序后,利用GenBank中蓝细菌不同科、属种类序列进行分子系统分析、系统发育树及遗传距离分析,结果表明测序样品应为Microcoleus vaginatusMicrocoleus streenstrupii

Abstract: In this paper, kinds of sand samples were isolated from Huairou, Beijing, using the method of 96-well plate limited dilution,and preliminarily identified as Synechococcus based on their morphological features respectively. ITS regions of these three samples was amplified, cloned to sequenced and finally blasted against the GenBank to compare with the homolog genes from the different genera in Synechococcus. The construction of phylogenetic tree and genetic distance analysis suggested the three samples may belong to Microcoleus vaginatus or Microcoleus streenstrupii.
